A nivel molecular, la respuesta de senescencia está sometida a una estricta regulación donde juegan un papel esencial genes y vías alterados en cáncer, como Retinoblastoma, p53 y los productos del locus CDKN2A, p16INK4A y ARF(Salama et al., 2014).

Las células senescentes se comunican con su entorno, principalmente mediante un conjunto de factores solubles liberados por estas células, que se conoce como el fenotipo secretor asociado a senescencia o SASP (Kuilman and Peeper, 2009). La acción del SASP es clave en la actividad de células senescentes in vivo, de maneras muy variadas. El SASP puede extender el fenotipo senescente a células de su entorno mediante un mecanismo paracrino y promover la eliminación de células senescentes mediante el reclutamiento de células del sistema inmune.

Pero paradójicamente, el SASP también tiene efectos perjudiciales, puesto que puede promover
el crecimiento tumoral o causar deterioro de los tejidos durante el envejecimiento. Esta dualidad de senescencia, con efectos tantos positivos como negativos tiene importantes implicaciones para el papel de las células senescentes in vivo.

Sin duda, medio siglo después del descubrimiento de Hayflick, el campo de senescencia vive un momento excitante y, por primera vez en todo este tiempo, quizás no esté muy lejos la fecha en que aquella observación del comportamiento de células en el laboratorio se pueda traducir en terapias efectivas para enfermedades humanas.
